Clinical Applications Research:

Turmeric (95% curcuminoids) is one of the most studied herbs for cognitive support (Planta Med. 2014 Oct;80(15):1249-58. Mechanisms of action of phytochemicals from medicinal herbs ....Kim MH, et al.)

Curcumin boosts DHA in the brain (BiochimBiophysActa. 2014 Dec 27;1852(5):951-961. Curcumin boosts DHA in the brain: Implications...Wu A, et al.) DHA is the long chain fatty acid that is essential for brain development and has been found in numerous clinical trials and observational studies to support cognitive function (Yurko-Mauro K. et al. Docosahexaenoic Acid ...: a systematic review and meta-analysis.PLoS One. 2015 Mar 18;10(3):e0120391). Curcumin demonstrated support for both nerve and glial cells (glial cells are non-neuronalcells that maintain homeostasis, form myelin, and provide support for neurons in the brain and peripheral nervous system) and axonal microenvironment (J Surg Res. 2015 Jan 7. pii: S0022-4804(15)00006-2. Curcumin...Yuan J, et al).

One review searched the literature (including PubMed, Science Direct, Scopus, and Cochrane Library) and found mechanisms of action to explain why the constituents of plants have support potential for neurotransmitters, including curcumin (Rev Neurosci. 2015 Feb 14. Plant-derived natural medicines for the management of depression: an overview of mechanisms of action.Farahani MS, et al). Curcumin from Curcuma longa interact with D1 and D2 receptors as well as elevate the brain dopamine level. Mechanism of support seems to be through Vitamin D receptors.

Turmeric (95% curcuminoids): Some scientists recognize the huge impact that a dietary component like curcumin can have: “The development of curcumin for clinical use as a regulator of epigenetic changes needs further investigation to determine novel and effective strategies, either alone or in combination with other agents, for improving outcomes” (Simone Reuter et al. Genes Nutr. 2011 May; 6(2): 93–108).

Ginkgo Biloba Extract (24% ginkgosides, 6% terpene lactones): Clinical trials report outcomes documenting brain support in patients (J Stroke Cerebrovasc Dis. 2013 Nov;22(8):e557-63. The effect of Ginkgo biloba...: a double-blind, placebo-controlled, randomized clinical trial.Oskouei DS1, et al). In an open clinical pilot study, GBE was administered to 20 children over 3 to 5 weeks. Ginkgo was found to have ideal characteristics to support the brain and behavior in children (Z Kinder JugendpsychiatrPsychother. 2014 Sep;42(5):337-47. Ginkgo biloba extract in children with ADHD.Uebel-von Sandersleben H. et al).

Ginkgo, along with phosphatidyl choline (as sunflower lecithin) and phosphatidyl serine are the most frequently recommended nutritional substances for cognitive support, “often in combinations with many other substances” (J Neuropsychiatry ClinNeurosci. 2010 Summer;22(3):345-7. A study of herbal remedies for memory complaints.Serby MJ, et. al).

Phosphatidyl Serine or phosphatidyl choline combined with ginkgo biloba appears to potentiate the effects of a low dose of ginkgo biloba (Hum Psychopharmacol. 2007 Jun;22(4):199-210. Acute cognitive effects of standardized Ginkgo biloba extract complexed with phosphatidylserine.Kennedy DO et al). The data suggest that combining standardized Ginkgo biloba extract (GBE) with soy-derived phospholipids enhances the bioavailability of GBE's active components. The current study therefore aimed to assess the comparative effects of a low dose of GBE with either phosphatidyl serine or phosphatidyl choline. The study utilized a placebo-controlled, multi-dose, double-blind, balanced-crossover design. Twenty-eight healthy young participants received either GBE, GBE with phosphatidylserine, or GBE with phosphatidylcholine or placebo. Ginkgo together with phosphatidylserine had the biggest effect, followed by ginkgo with phosphatidyl choline.

We have always recognized the essential role for choline. Choline (as bitartrate) is a necessary element and building block for the structural components of cell membranes and helps account for fluidity and communication within those membranes. Choline is essential for phosphatidyl choline and sphingomyelin, which are the two major phospholipids in cell walls. Choline is the precursor molecule for the neurotransmitter acetylcholine, which is involved in many functions of the brain and muscles.

Choline has many forms, several of which have been called “designer cholines” because they exhibit properties above and beyond regular choline. Two of these designer cholines are known as alpha-GPC, or Glycerolphosphorylcholine, and CDP choline, or Cytidine 5'-diphosphocholine.

Additionally, phosphtidyl choline and phosphatidyl serine have been discovered as nutritional additions to help boost the absorptive power of other ingredients that are given in conjunction with them, such as ginkgo and curcumin for instance (Drug Deliv. 2014 Nov 4:1-9. Curcumin-loaded nanostructured lipid carriers (NLCs) for nasal administration: design, characterization, and in vivo study.Madane RG, Mahajan HS.; Hum Psychopharmacol. 2007 Jun;22(4):199-210. Acute effects of standardized Ginkgo biloba extract complexed with phosphatidylserine.Kennedy DO et al.; Planta Med. 2001 Jun;67(4):326-30. Complexation of Ginkgo biloba extract with phosphatidylcholine improves activity and increases the plasma antioxidant capacity in the rat.Carini M, et al.; Phytother Res. 2006 Oct;20(10):901-5.Neuropharmacological evaluation of Ginkgo bilobaphytosomes...Naik SR, et al.)

Alpha glycerolphosphorylcholine (Alpha GPC demonstrated a support benefit for patients in a number of clinical studies conducted on the brain and maintenance of brain function. In fact, the researchers, “being impressed not only with results but in lack of side effects, recommended that it is desirable to reconsider alpha glycerolphosphorylcholine in larger carefully controlled studies ... for the maintenance of brain function” (Scapicchio PL. Revisiting choline alphoscerate profile: a new, perspective, role...Int J Neurosci. 2013 Jul;123(7):444-9.)

“Supplementation with alpha-GPC and CDP choline (cytidine 5'-diphosphocholine) has proven to be much superior to supplementation with just basic choline for brain support and normal neurotransmitter production. It is theorized that these designer cholines are able to be better incorporated and stored into the phospholipids in the brain, and therefore make better precursors for acetylcholine” - Amenta F, Tayebati SK. Pathways of acetylcholine synthesis, transport and release. Curr Med Chem. 2008;15(5):488-98.

More on Alpha GPC:

A multicenter, randomized, controlled study compared the efficacy (for brain support) of alpha-GPC among 126 patients, and found that alpha GPC provides excellent support (Drugs Aging. 1993 Mar-Apr;3(2):159-64.Multicentre study of l- alpha-glyceryl-phosphorylcholine. Parnetti L. et al).

“Alpha- glycerolphosphorylcholine, (alpha-GPC) is a derivative of phosphatidyl choline known to support the parasympathetic nervous system. Alpha-GPC demonstrated benefit for patients in a number of clinical studies conducted on the brain and maintenance of brain function. The researchers, being impressed not only with results but in lack of side effects, recommended that “it is desirable to reconsider alpha-GPC in larger carefully controlled studies”, ... for the maintenance of brain function. Scapicchio PL. Revisiting choline alphoscerate profile: a new, perspective, role...?Int J Neurosci. 2013 Jul;123(7):444-9. CDP Choline undergoes quick hydrolysis and dephosphorylation after ingestion to yield cytidine and choline, which then enter the brain separately and are used to resynthesize CDP-choline inside brain cells. A research paper reviews the roles of choline and of choline-containing phospholipidson brain metabolism, stating that (CCPLs) are safe, effective compounds developed a long time ago thathave a place in this support category and should be further investigated by new appropriate clinical studies. They also report that CDP-choline has displayed interesting effects documented in preclinical studies and limited clinical trials for its neurotransmission support (ClinChem Lab Med. 2013 Mar 1;51(3):513-21. Choline-containing phospholipids: relevance to brain functional pathways.Tayebati SK1, Amenta F.) In an open, multicenter trial, CDP choline supported biosynthesis of phospholipids in neuronal membranes, brain metabolism, neurotransmitters, and the central nervous system (ClinInterv Aging. 2013;8:131-7. Effectiveness and safety of citicoline...: the IDEALE study.Cotroneo AM1, et al).
